| 内容記述 | The effective dielectric constants of protoplasts which are proportional to dielectrophoretic force are investigated by impedance measurements. Daucas carota is used as a plant in this experiment. Protoplasts are washed and suspended in sucrose solution. The spectrum of conductivity and that of relative dielectric constants for both a suspension of protoplasts and a suspending medium are measured, respectively. Then, the spectra of conductivity, relative dielectric constants and effective dielectric constants for a protoplast are calculated in the range of 100kHz to 10 MHz. The spectrum of effective dielectric constants shows one peak. The peak is considered to be due to the difference between relative dielectric constant of suspending medium and that of protoplast. In addition, the single shell model of a cell that consists of a membrane and an interior is used. The conductivity and the relative dielectric constant for both a membrane and a interior using a curve fitting method to the spectrum of complex dielectric constants of a protoplast, are calculated. Also, the spectra of effective dielectric constant are obtained as a function of conductivity of a suspending medium. The frequency range of positive dielectrophoretic force is from about 200 kHz to about 400 MHz for a conductivity of 100 S/cm. It is found that a suspending medium of low conductivity is suitable for dielectrophoretic manipulation of protoplasts. | |||||
