However, AldDH is not studied well in a view of ω-oxidation pathway. We found a novel AldDH specific for ω-oxo fatty acids in rabbit liver cytosolic fraction. This paper describes the purification of AldDH from rabbit cytosolic fraction, the cloning of a cDNA encoding the AldDH and its expression in E. coli. An AldDH specific for 12-oxo lauric acid was purified to a single band, which showed an apparent molecular weight of 50 kDa on SDS-PAGE. A cDNA (2.1 kbp) encoded a protein of 500 amino acids has been isolated from rabbit liver cDNA library. The amino acid sequence predicted from the cDNA showed 90% identity with that of human retinal dehyderogenase. Purified AldDH (AldDH-C) and AldDH expressed in E. coli (AldDH-R) recognize as substrates 16-oxo palmitic acid, 12-oxo lauric acid, 10-oxo capric acid, and 8-oxo caprylic acid. 12-oxo lauric acid dehydrogenase activity in rabbit liver cytosolic fraction was inhibited by an antibody against AldDH-R with a same manner of AlcDH-R.
