The alkaline phosphatase is synthesized in response to glycerophosphate starvation and is ion strength-dependent. The phosphatase has been purified to homogeneity from glycerophosphate-starved thalli. The phosphatase is comprised of two subunits, each having an apparent molecular mass of 76 kDa by SDS-polyacrylamide gel electrophoresis. A cDNA clone of the alkaline phosphatase has been isolated by a combination of 3' rapid amplification of cDNA ends and oligo-capping method from thalli grown under glycerophosphate-deficient conditions. The cDNA contained a 2,293 bp open reading frame encoding 29 residues for signal peptide and 658 residues for the mature form of phosphatase.
